If Mikrotik software supports it, yes. I believe most third party routerOS only has the functionality if at most is OpenVPN client capability meaning that you can only use OpenVPN sort of setup to authenticate with any of the VPNGate servers.
From what I know of, SoftEther VPN has been tried mainly on machines other than embedded platforms such as routers, to make SoftEther VPN work on routers would require one the knowledge to cross-compile and to basically address issues as they arise as cross-compiling isn't always a simple affair.
You are probably better off using your own physical computer for VPNGate servers and not your router considering that VPNGate servers are mainly donated by volunteers whom can do whatever they want with their connection. OpenVPN connectivity on router may get really messy with VPNGate servers as you are not only gambling your luck with the poor volunteer but also errors that are maybe difficult to diagnose. Both SoftEther VPN and OpenVPN assumes the connection to the VPN host is stable, if it is not stable it assumes the user would be able to quickly discover and rectify any issues upon the loss of connectivity with the VPN host. Failing that would obviously lead to the client ending up to use their own connection and not a VPN connection.
