I will be direct here.
I have an openvpn server configured, located at Singapore using Debian Linux.
Now, I have a windows OS based pc.
Normally, I connect to my openvpn server by using OpenVPN GUI (with .ovpn config file).
I am using certificate based authentication.

tls-auth
Any idea on how to adapt it for SoftEther Client?

Simply, there is no way. SoftEther client can't "speak" ovpn. 
You can use a OpenVPN Client to conect to a SoftEther server but not vice versa. You can install SoftEther on the server and use both on client side, OpenVPN and SoftEther Client
